Mixed depressant and stimulant actions of morphine and their relationship to brain acetylcholine.

چکیده
Morphine and related narcotic agonists produce mixed depressant and/or stimulant effects in a wide variety of animals including man (1,2) . These effects are not only species but also time and dose related . It is not gener ally appreciated that even a single dose of morphine in a given species, such as the rat, may have initial depressant and subsequent stimulant effects. We believe this is an extremely important concept and intend to provide evidence of its wide applicability in the pharmacology of narcotics .

The Preliminary Excitant Action of Morphia
Sir,?In man, morphine in therapeutic doses, has a depressant action on the central nervous system, though in some animals it has a transient stimulant action? especially on the spinal cord. But even in animals this stimulant action is limited to the spinal cord; and the brain is depressed without any preliminary phase of stimulation. I was therefore surprised when, some time ago, I gave a hypod...
متن کاملPhysostigmine antagonizes morphine-induced respiratory depression but not analgesia in dogs and rabbits.
The ability of physostigmine to antagonize the respiratory depressant effect of morphine was studied in conscious rabbits and ketamine-anaesthetized dogs pretreated with atropine methyl nitrate. Morphine 4 mg kg-1 increased PaCO2 in the rabbit from 3.43 +/- 0.16 to 4.95 +/- 0.28 kPa, decreased arterial pH from 7.45 +/- 0.01 to 7.31 +/- 0.01 and decreased respiratory frequency by 36%. Physostigm...
متن کاملMorphine-Induced Place Preference: Interactions with Neurotransmitter Systems
This review gives an overview of our recent findings and developments in research on brain mechanisms of morphine reward from studies using the place preference conditioning paradigm. Intracranial place conditioning methodology has become a valuable and firmly established and very widely used tool in behavioural pharmacology and drug reward mechanisms. Several studies have established that morp...

متن کاملThe Effect of Intra-Medial Septum Co-Injection of Ghrelin and Nicotine on Morphine Induced Memory Impairment
Evidence indicates that medial septum nicotinic receptors regulate cognitive process. Ghrelin is a gut hormone that regulates energy homeostasis. Ghrelin also is produced in brain and known to be involved in cognitive function. The purpose of the study was to evaluate the effects of medial septal injection of ghrelin on morphine-induced memory impairment in rats. Also, the possible relationship...
